Methods in Toxicology, Volume 2: Mitochondrial Dysfunction provides a source of methods, techniques, and experimental approaches for studying the role of abnormal mitochondrial function in cell injury. The book discusses the methods for the preparation and basic functional assessment of mitochondria from liver, kidney, muscle, and brain; the methods for assessing mitochondrial dysfunction in vivo and in intact organs; and the structural aspects of mitochondrial dysfunction are addressed. The text also describes chemical detoxification and metabolism as well as specific metabolic reactions that are especially important targets or indicators of damage. The methods for measurement of alterations in fatty acid and phospholipid metabolism and for the analysis and manipulation of oxidative injury and antioxidant systems are also considered. The book further tackles additional methods on mitochondrial energetics and transport processes; approaches for assessing impaired function of mitochondria; and genetic and developmental aspects of mitochondrial disease and toxicology. The text also looks into mitochondrial DNA synthesis, covalent binding to mitochondrial DNA, DNA repair, and mitochondrial dysfunction in the context of developing individuals and cellular differentiation. Microbiologists, toxicologists, biochemists, and molecular pharmacologists will find the book invaluable.

This volume describes a broad spectrum of experimental approaches for investigating structure, function, and transport of neuronal mitochondria in health and disease. Most of these approaches were only recently developed and range from electron tomography-based 3D reconstruction of mitochondrial cristae to patch clamp recording from mitochondria in intact neurons. The chapters in this book cover topics such as mitochondrial proteomics, fluorescence lifetime imaging, respirometry and mitophagy, as well as optical approaches based on the use of genetically engineered fluorescent sensors for monitoring synaptic ATP and axonal ROS generation, mitochondrial Ca2+ cycling and pH changes, and mitochondrial dynamics and axonal trafficking in live neurons. Each chapter also discusses difficulties, tips, tricks, and precautions to take. Neuromethods series style chapters include the kind of detail and key advice from the specialists needed to get successful results in your laboratory. Cutting-edge and comprehensive, Techniques to Investigate Mitochondrial Function in Neurons is a valuable and useful resource for a broad range of investigators interested in the function of neuronal mitochondria in health and disease states.

These are exciting times for the field of optical imaging of brain function. Rapid developments in theory and technology continue to considerably advance understanding of brain function. Reflecting changes in the field during the past five years, the second edition of In Vivo Optical Imaging of Brain Function describes state-of-the-art techniques and their applications for the growing field of functional imaging in the live brain using optical imaging techniques. New in the Second Edition: Voltage-sensitive dyes imaging in awake behaving animals Imaging based on genetically encoded probes Imaging of mitochondrial auto-fluorescence as a tool for cortical mapping Using pH-sensitive dyes for functional mapping Modulated imaging Calcium imaging of neuronal activity using 2-photon microscopy Fourier approach to optical imaging Fully updated chapters from the first edition Leading Authorities Explore the Latest Techniques Updated to reflect continuous development in this emerging research area, this new edition, as with the original, reaches across disciplines to review a variety of non-invasive optical techniques used to study activity in the living brain. Leading authorities from such diverse areas as biophysics, neuroscience, and cognitive science present a host of perspectives that range from a single neuron to large assemblies of millions of neurons, captured at various temporal and spatial resolutions. Introducing techniques that were not available just a few years ago, the authors describe the theory, setup, analytical methods, and examples that highlight the advantages of each particular method.

Primary sensory neurons respond to peripheral stimulation and project to the spinal cord. Specifically, the population of neurons which respond to damaging stimuli terminate in the superficial layers of the dorsal horn. Therefore, the dorsal horns constitute the first relay site for nociceptive fibre terminals which make synaptic contacts with second order neurons. It has recently become clear that the strength of this first pain synapse is plastic and modifiable by several modulators, including neuronal and non-neuronal regulators, and studies on the fundamental processes regulating the plasticity of the first pain synapse have resulted in the identification of new targets for the treatment of chronic pain. This book will be of interest to a wide readership in the pain field.
