Rapid progress in the field of organophosphate compounds has made this new edition necessary. Particular attention has been paid to new developments in biologically active products as this is probably of greatest interest to practical chemists. I have therefore updated the chapter on chemistry and, in connection with this, rewritten the chapter on metabolism. Elberfeld, January 1982 CHRISTA FEST VII Preface to the First Edition Our intention has been to provide a short introduction to the chemistry and mode of action of insecticidal phosphoric acid compounds, with particular ref erence to the relationship between structure and activity. The yearly produc tion of these pesticides is now approaching 100,000 tons and thus offers an im portant example of applied research. If, however, one examines the historical development of these compounds, it is apparent that this was preceded by a hundred years of pure chemistry of phosphorus.

This book aims to open new fields of interest in phosphate chemistry and to suggest a new system of classification for phosphates. Due to the very rapid development of this branch of chemistry during the last thirty years and the very confusing nomenclature often used in chemical literature, this first attempt seems not only justified but necessary. The suggested classification is not revolutionary, with respect to condensed phosphates for instance, but establishes clear boundaries between some categories of compounds such as adducts and heteropolyanion-compounds. The first chapters cover the present state of phosphate chemistry in a general way and could serve as an introduction to phosphates for some categories of students or teachers. The next few chapters are devoted to more specialized topics which would be of interest to chemists, crystallographers or solid-state specialists.Some specific aspects of the crystal chemistry of phosphates, such as the geometry of the networks built by acidic phosphoric anions, for instance, are covered for the first time. The final chapter suggests some improvements for the present system of classification for phosphates.In addition, presented for the first time are the results recently obtained for the replacement of asbestos by harmless biodegradable phosphate fibers.

Undoubtedly the chemistry of phosphorus appears more and more attractive in the sense that phosphorus presents an extraordinary versatile behaviour that allows the synthesis of a large number of different phosphorus reagents usable for applications in different fields, from biology to material science without f- getting key applications in catalysis. The full maturity of this topic of research can be explained by all the acquired knowledge over these twenty last years. Organic and inorganic chemistries of P , P and P species have been the s- in ]V v ject of thousands of publications while the chemistry of low coordinated ph- phorus derivatives experienced its hour of glory from the 1970s to the early 1990s. In parallel, interactions between phosphorus compounds and transition metals afforded many complexes, a lot of which present fascinating properties as catalysts. The interest in all these themes really is not decreasing, indeed some fas- nating areas of research are emerging or are the subject of many investigations: the medicinal chemistry of bisphosphonates, the role of phosphorus in biology, phosphorus ligands in radiopharmaceutical chemistry, phosphorus in material science, new polymers and dendrimers incorporating phosphorus, and as- metric catalysis to name but a few. Indeed researchers, benefitting from their background in basic phosphorus chemistry are developing many new fields of research.

The first two chapters of this invaluable book trace the developments of the chemistry and macromolecular structures, respectively, of proteins and nuclei acids. Similarly, the introductions to the succeeding chapters review, step by step, the historical landmarks in the topics covered. These include discoveries of biological phosphate esters, nucleotides and nucleotide coenzymes (important in intermediary metabolism), the nature of the genetic material and biological synthesis of proteins, formulation of the problem of the genetic code, and perspectives on bioenergetics.The selected papers illustrate the developments of the chemical synthesis of nucleotides and nucleotide coenzymes of ribo- and deoxy-ribo-polynucleotides (RNA, DNA), of the total synthesis of genes in the laboratory, and principles for gene amplification (PCR). Another major section covers studies of enzymes that degrade nucleic acids, the structure of transfer RNA and its role in protein synthesis, and the author's work on the elucidation of the genetic code. Finally, there are descriptions of the studies on biological membranes and the membrane protein bacteriorhodopsin, a biological proton pump. These studies elucidated the mechanism of proton translocation, which is central to bioenergetics.
