This CTR-Special is on one of the most exciting topics in the neurosciences, the regulation of axonal growth during development and in the injured nervous system by attraction and repulsion. How the specific wiring of billions of neuronal connections is achieved in the brain has been of interest since the days of Cajal, and even earlier. The implications of this topic extend far beyond developmental neurobiology into the fields of applied medicine, clinical neurology and related disciplines. Following the discoveries of Eph receptors and their ligands, semaphorins and netrins, the field has greatly expanded in the past 2 years and continues to grow. Much of the current fascination for some of these molecules is related to their ambiguous roles in axonal growth promotion and repulsion.

Axon Growth and Regeneration: Methods and Protocols brings together a diverse set of techniques for the study of the mechanisms underlying central nervous system axon growth, consequently providing a resource that will aid in the development of repair strategies. After an introductory section, this detailed volume continues with sections focusing on axon growth in vitro, providing a range of protocols that can be used to examine intracellular signalling pathways, axonal responses to extracellular factors and methods for quantifying outgrowth. The next section provides protocols for inducing experimental injury in vivo as well as some highly promising protocols for promoting regeneration, which segues into the final section highlighting a series of protocols that can be used to monitor the extent of axon regeneration in vivo, ranging from tract tracing to in vivo imaging and functional recovery. As a book in the Methods in Molecular Biology series, chapters contain introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols and tips on troubleshooting and avoiding known pitfalls. Practical and reliable, Axon Growth and Regeneration: Methods and Protocols aims to serve researchers studying axon regeneration with a significant set of diverse tools, vital for moving on to the next generation of exciting new discoveries in the field.

The genetic, molecular, and cellular mechanisms of neural development are essential for understanding evolution and disorders of neural systems. Recent advances in genetic, molecular, and cell biological methods have generated a massive increase in new information, but there is a paucity of comprehensive and up-to-date syntheses, references, and historical perspectives on this important subject. The Comprehensive Developmental Neuroscience series is designed to fill this gap, offering the most thorough coverage of this field on the market today and addressing all aspects of how the nervous system and its components develop. Particular attention is paid to the effects of abnormal development and on new psychiatric/neurological treatments being developed based on our increased understanding of developmental mechanisms. Each volume in the series consists of review style articles that average 15-20pp and feature numerous illustrations and full references. Volume 2 offers 56 high level articles devoted mainly to Formation of Axons and Dendrites, Migration, Synaptogenesis, Developmental Sequences in the Maturation of Intrinsic and Synapse Driven Patterns. - Series offers 144 articles for 2904 full color pages addressing ways in which the nervous system and its components develop - Features leading experts in various subfields as Section Editors and article Authors - All articles peer reviewed by Section Editors to ensure accuracy, thoroughness, and scholarship - Volume 2 sections include coverage of mechanisms which regulate: the formation of axons and dendrites, cell migration, synapse formation and maintenance during development, and neural activity, from cell-intrinsic maturation to early correlated patterns of activity

In this treatise, current knowledge on the neurogenesis, axonal outgrowth, synaptogenesis, and regenerative capacity of descending supraspinal pathways in tetrapods is discussed. Although emphasis is on the clawed toad, Xenopus laevis, chicken embryos, opossums and rodent data, also the data available for primates including man are presented. It will be shown that 1) the outgrowth of descending supraspinal pathways is the result of a coordinated program; 2) the pattern of early descending axonal tracts is similar in all vertebrate groups; 3) the formation of descending supraspinal pathways occurs according to a developmental sequence; 4) the earliest descending supraspinal fibers arrive in a rather immature spinal cord, and 5) the regenerative capacity of descending supraspinal pathways depends on the developmental stage the particular pathways arise.

This volume addresses central issues in the embryological development of the nervous system, which, while similar to that of other organs and tissues, features special and intricate mechanisms of morphogenesis. It emphasizes the description of cellular interactions at the molecular level and includes some of the first fruits from applications to neurobiology of recent developments in othera areas--notable immunology, biochemistry, molecular biology, and molecular genetics.
