White matter development in the human brain undergoes a uniquely extended developmental trajectory, and the maturation of this complex network of connections is broadly relatable to real-world measures of cognitive ability. We begin this dissertation with a review of the literature on structural brain development, in order to provide a useful background layer for our discussion (Chapter 1). Next, we report a case study investigating white matter abnormalities in the context of prenatal methamphetamine exposure, and their cognitive correlates (Chapter 2). Then we describe our successfully-funded NIH grant proposal to 1) develop novel image analysis techniques for investigating white matter connectivity, and 2) apply them to the study of a) typical frontal lobe white matter maturation, b) its relation to executive functioning, and c) how these processes are affected by prenatal alcohol exposure (Chapter 3). The results of this effort are described in Chapter 4, Chapter 5, and Chapter 6. Finally, we supplement the work on our UCLA cohort with exciting results from two large, multi-site, collaborative efforts: 1) ADHD-200, an initiative to employ brain mapping findings in a machine learning environment for the diagnostic classification of individual subjects with attention deficit hyperactivity disorder (ADHD) (Chapter 7), and 2) PING (the Pediatric Imaging, Neurocognition, and Genetics study), where we are leading an effort to provide the most comprehensive mapping to-date of typical white matter development (Chapter 8).

An up-to-date overview of the field of connectomics, introducing concepts and mechanisms underlying brain network change at different stages. The human brain undergoes massive changes during its development, from early childhood and the teenage years to adulthood and old age. Across a wide range of species, from C. elegans and fruit flies to mice, monkeys, and humans, information about brain connectivity (connectomes) at different stages is now becoming available. New approaches in network neuroscience can be used to analyze the topological, spatial, and dynamical organization of such connectomes. In Changing Connectomes, Marcus Kaiser provides an up-to-date overview of the field of connectomics and introduces concepts and mechanisms underlying brain network changes during evolution and development. Drawing on a range of results from experimental, clinical, and computational studies, Kaiser describes changes during healthy brain maturation and during brain network disorders (including such neurodevelopmental conditions as schizophrenia and depression), brain injury, and neurodegenerative disorders including dementia. He argues that brain stimulation is an area where understanding connectome development could help in assessing long-term effects of interventions. Brain health affects Americans across all ages, genders, races, and ethnicities. Enriching the body of scientific knowledge around brain health and cognitive ability has the potential to improve quality of life and longevity for many millions of Americans and their families. The Centers for Disease Control and Prevention estimate that as many as 5 million Americans were living with Alzheimer's disease in 2014. That same year, more than 800,000 children were treated for concussion or traumatic brain injuries in U.S. emergency departments. Each year, more than 795,000 people in the United States have a stroke. Developing more effective treatment strategies for brain injuries and illnesses is essential, but brain health is not focused exclusively on disease, disorders, and vulnerability. It is equally important to better understand the ways our brains grow, learn, adapt, and heal. Addressing all of these domains to optimize brain health will require consideration about how to define brain health and resilience and about how to identify key elements to measure those concepts. Understanding the interactions between the brain, the body, and socioenvironmental forces is also fundamental to improving brain health. To explore issues related to brain health throughout the life span, from birth through old age, a public workshop titled Brain Health Across the Life Span was convened on September 24 and 25, 2019, by the Board on Population Health and Public Health Practice in the Health and Medicine Division of the National Academies. This publication summarizes the presentation and discussion of the workshop.
