This work covers the pathophysiology of cancer, exploring the difficulty of optimal treatment due to the complexity and diversity of cancer types. The search for distinctive molecular biology characteristics of tumor cells is especially relevant in the identification of overexpressed receptors and proteins that can be used as a target for cancer treatment. We highlight the main therapeutic modalities, particularly conventional systemic chemotherapy, addressing its mechanisms of action, therapeutic classes and even the toxic effects. We also describe the main tumor markers, their importance in the diagnosis and treatment of cancer, and the specificity of tumor cells. The first chapters serve as an introduction to the central topic of this book, targeted therapy. Key aspects of target therapy, such as classes of drugs, immunotherapy, monoclonal antibodies, checkpoint inhibitors, cancer vaccines and tyrosine kinase inhibitors are presented, and, for each one, the benefits, as well as the adverse effects are reported. Chapter 6 compares conventional systemic chemotherapy and targeted therapy, identifies the risks and benefits and also the eligibility criteria for patient care. The possibility of targeted therapy replacing conventional chemotherapy is discussed while reviewing studies that demonstrate the benefits of combining both types of treatment. Finally, the introduction of pharmaceutical nanotechnology to improve antineoplastic agents is addressed in the last chapter and sets the direction for future research in cancer treatment. This is a valuable resource for many health professionals including physicians, pharmacists, nurses, researchers and students interested in the field of oncology.

The stimulation of the immune system through artificial means for the treatment of cancer is known as cancer immunotherapy. It boosts the ability of the immune system to fight cancer cells. It is a subspecialty within oncology. The antibody proteins belonging to the immune system are able to detect the tumor antigens and bind with them. Immunotherapy can be broadly divided into two types, namely, passive immunotherapy and active immunotherapy. The tumor cells are specifically targeted through the immune system in active immunotherapy. Within passive immunotherapy, the cancer cells are not directly attacked by the immunotherapy, but the ability of the immune system to attack cancer cells is enhanced. This book includes some of the vital pieces of work being conducted across the world, on various topics related to cancer immunotherapy. Also included herein is a detailed explanation of the various concepts and applications of this branch of therapy. This book includes contributions of experts and scientists which will provide innovative insights into this field.

The field of immuno-oncology continues to rapidly evolve as new insights to fight and treat cancer emerge. The fourth edition of Immunotherapy provides the most current overview of immuno-oncology in different cancer types and toxicities associated with immunotherapy. While immunotherapy has revolutionized the treatment landscape of several solid malignancies, several challenges still exist. Only a subset of patients derive clinical benefits; some do not respond at all, and others respond initially, only for their disease to progress later. Because these drugs can activate a broad range of immune cells, patients suffer from a unique set of side effects known as immune-related adverse events. As more immunotherapeutic agents are used in the clinic, it is important to provide updates about current and ongoing developments in the field to further research efforts and inform treatment decisions. The fourth edition will have a new focus on strategies to overcome the challenges associated with immunotherapy. Chapters will discuss topics such as biomarkers of response, resistance mechanisms, role of imaging in predicting immune-related adverse events, and management of immune-related adverse events. Written by leading experts conducting cutting-edge research, readers will gain up-to-date knowledge on the current state and future of immunotherapy.

This timely book, published just as cancer immunotherapy comes of age, summarizes the rationale, present status, and future perspective for cancer immunotherapy. Included are explanations of the constitution of the immune system and immunocheckpoints, the mechanism of antigen presentation and recognition, valuable modalities, clinical trials and guidance, personalization, and biomarkers, all of which are essential for understanding the success of cancer immunotherapy. This innovative therapy has been investigated worldwide as the fourth line of cancer treatment after the standard treatments of surgery, chemotherapy, and radiotherapy. The progress in fundamental understanding of tumor immunology and the recent advances in clinical trials have opened new avenues with a cancer vaccine in 2010 and immunocheckpoint modulation in 2011, with their approval already granted in the United States. Today, there are no doubts, even among experts in cancer chemotherapy and radiotherapy, that the immune system plays a vital role in tumor eradication. Following American approval, many clinical trials of cancer immunotherapy are being conducted. With this book the reader will readily understand the paradigm shift in cancer treatment and will realize the importance of cancer immunotherapy. The great value of immunotherapy will be obvious, not only for tumor shrinkage but for prolonging patient survival.
