Various metabolic functions associated with the Escherichia coli membrane system were analyzed in mutant strains lacking adenylate cyclase (cya mutations) or cyclic AMP binding protein (CRP) activity (crp mutations). Mutations which have suppressed the organism's requirement for cyclic AMP were also studied, and mutants of Salmonella typhimurium were constructed which are cya or crp in addition to lacking the enzymes of the phosphoenolpyruvate: phosphotransferase system (PTS). These studies have established that key transport and bioenergetic activities of the enterobacterial membrane are regulated at the genetic level of transcription by the cAMP-CRP regulatory complex. The ability of E. coli to transport substrates via PTS and non-PTS routes are under this control. The organism's ability to generate a proton motive force (PMF) and to link this force to transport activities and ATP synthesis are also under this control. All parts of the E. coli envelope, the inner, cytoplasmic membrane, the periplasmic region, and the outer membrane are to one degree or another affected by the cAMP-CRP regulatory function.

Interfacial Phenomena examines the fundamental properties of various liquid interfaces. This book discusses the physics of surfaces; electrostatic and electrokinetic phenomena; and adsorption at liquid interfaces. The properties of monolayers; reactions at liquid surfaces; diffusion through interfaces; and disperse systems and adhesion are also deliberated. Other topics include the vapor pressures over curved surfaces; electrical capacity of the double layer; applications of electrophoresis; and thermodynamics of adsorption and desorption. The experimental methods of spreading films at the oil-water interface; penetration into monolayers; experiments on dynamic systems; and spontaneous emulsification are likewise covered in this text. This book is beneficial to chemical engineers and students concerned with interfacial phenomena.

This volume is comprised of a collection of experimental protocols for common techniques and strategies used to study the biogenesis of b-barrel outer membrane proteins in Gram-negative bacteria. The BAM Complex: Methods and Protocols guides readers through methods on the function of the BAM complex, the roles played by each of the individual components, the expression and purification of the components, crystallization and structure determination of the components, and how the individual Bam components may assemble into a functional complex. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Authoritative and cutting-edge, The BAM Complex: Methods and Protocols will serve as an invaluable reference for those interested in studying the BAM complex.
