This primary textbook for a first course in pharmacology offers an integrated, systems-based, and mechanism-based approach to understanding drug therapy. Each chapter focuses on a target organ system, begins with a clinical case, and incorporates cell biology, biochemistry, physiology, and pathophysiology to explain how and why different drug classes are effective for diseases in that organ system. Over 400 two-color illustrations show molecular, cellular, biochemical, and pathophysiologic processes underlying diseases and depict targets of drug therapy. Each Second Edition chapter includes a drug summary table presenting mechanism, clinical applications, adverse effects, contraindications, and therapeutic considerations. New chapters explain how drugs produce adverse effects and describe the life cycle of drug development. The fully searchable online text and an image bank are available on thePoint.

Thoroughly updated and completely reorganized for a sharper clinical focus, the Fifth Edition of this world-renowned classic synthesizes the latest advances in basic neurobiology, biological psychiatry, and clinical neuropsychopharmacology. The book establishes a critical bridge connecting new discoveries in molecular and cellular biology, genetics, and neuroimaging with the etiology, diagnosis, and treatment of all neuropsychiatric disorders. Nine sections focus on specific groups of disorders, covering clinical course, genetics, neurobiology, neuroimaging, and current and emerging therapeutics. Four sections cover neurotransmitter and signal transduction, emerging methods in molecular biology and genetics, emerging imaging technologies and their psychiatric applications, and drug discovery and evaluation. Lithium, the treatment of choice for mania since its usefulness was first reported, has been shown to have varied effects on multiple biological systems, including electrolyte flux and neurochemistry. Recent advances in cellular and molecular biology promise to provide clinicians with a better understanding of the etiology of bipolar disorder and new options for treatment. Bipolar Medications: Mechanisms of Action presents the treatment and prophylaxis of bipolar disorder. More than 40 investigators share research and insight into the neurobiological mechanisms that help to explain the powerful effects of new antibipolar drugs. This comprehensive text Examines valproic acid, lamotrigine, inositol monophosphatase inhibitors, and protein kinase C inhibitors that have the potential to revolutionize clinical practice and provide new hypotheses on the etiology of bipolar disorder Presents the current understanding of the cellular mechanisms of action of mood-stabilizing agents Discusses the emergence of valproate as a powerful lithium alternative and examines the preliminary indications that lamotrigine will be an effective option Examines the issue of withdrawal rebound, which can make lithium ineffective or even counterproductive, and reviews inositol monophosphatase inhibitors that mimic lithium action in patients Compares lithium, carbamazepine, and valproate and their differential mechanisms, which could form the basis of a "rational polypharmacy" of manic depressive illness Examines behavioral models important in the screening of new antibipolar compounds and the effects of antibipolar compounds on immediate early genes Complete with extensive references, tables, and figures, this text is essential reading for any clinician who treats patients with bipolar disorder. It thoroughly documents the latest in psychopharmacology, as well as projecting future advances in the treatment of bipolar disorder.

The Editorial Board and the Publishers of the Handbook of Experimental Pharmacology wish to express their profound grief at the untimely death of Professor Peter Baker. Aware of his international recognition as an expert on the ubiquitous role of calcium in physiological processes and their pharma cological control, the Board was gratified when Professor Baker accepted its invitation to edit a new Handbook volume on "Calcium in Drug Actions". He went about this task with his usual energy and effectiveness so that, in the few months before his unexpected death, Professor Baker had mustered his distinguished contributors, got them to provide their manuscripts, and seen almost the entire material into the press. This achievement is all the more remarkable when one bears in mind the extraordinary number of his other commitments during the same time; they are mentioned in Sir Alan Hodgkin's preface to this volume. With so many other professional and personal responsibilities upon him, the Board of the Handbook wishes to record its grateful appreciation for the admirable way in which Professor Baker took on and carried out the additional work of bringing this fine book into existence; and the Board wishes it to be dedicated to the memory of Professor Peter Frederick Baker. This book is presented as a 1989 update on the task set by Robert Burton in his "Anatomy ofMelancholy," published in 1621. Burton's treatise addressed ques tions regarding depression which are still highly relevant today: ." . . What is it, with all the kinds, causes, symptoms, prognostickes and several cures ofit. . . . " These remain the core issues in affective disorders notwithstanding the remarkable progress that has been made in addressing them. New Directions in Affective Disorders sets out to provide an overviewofwhat has been achieved with particular emphasison developing trends and novel initiatives in bothfundamental research and treatment. The overriding objective of the book is to integrate significant contributions from basic and clinical science into a comprehensive format which will be of value to both clinicians and researchers. Intensive interest in affective illness isan inevitableconsequenceofthe frequency with which these disorders occur. Depression is the most common psychiatric condition, affecting as many as 50%ofpeople in Western countries at some time in their lives. National Institute ofMental Health Statistics estimate that 15% of adults between 18 and 74 years ofage may be suffering from serious depressive disorders in any given year. Depression is a serious condition with a high mortality. A suicide rate ofapproximately 20,000 deaths per year as a result of this illness is almost certainly a conservative estimate."
