Breaking protocol is about God bypassing procedures, established processes and natural laws for His will in one's life. It is God doing things that defy all hu - man logic, things that are unthinkable, unnatural, unusual, unparalleled, and unprecedented. You may be going through a season of no form, no light, no hope, no peace, no joy or however you choose to describe your situation but know that God is still in the business of breaking natural laws, human laws and even spiritual laws for His name sake. He has done it before, and He can do it again. He is the unchangeable changer. He is not limited and cannot be constrained by the circumstances of your life. He is the same yesterday, today and forever. Jesus Christ the same yesterday, and today, and forever. (Hebrews 13:8) For I am the Lord, I change not; therefore, ye sons of Jacob are not consumed. (Malachi 3:6). God will never allow you to be consumed; that situation is not meant to see your end. It is to glorify God; as Jesus declared to the sisters of Lazarus when they sent a message to him that their brother Lazarus whom He loved was sick. When Jesus heard that, he said, this sickness is not unto death, but for the glory of God, that the Son of God might be glorified thereby. (John 11:4). In fulfillment of His promise, in verse 43-44 of the same chapter, Jesus went ahead to break protocol for Lazarus' sake. As you read this book, desire a change, open up your spirit, and get insight into how you can properly position yourself for a "Breaking Protocol Experience". The first edition of Protein Purification Protocols (1996), edited by Professor Shawn Doonan, rapidly became very successful. Professor Doonan achieved his aims of p- ducing a list of protocols that were invaluable to newcomers in protein purification and of significant benefit to established practitioners. Each chapter was written by an ex- rienced expert in the field. In the intervening time, a number of advances have w- ranted a second edition. However, in attempting to encompass the recent developments in several areas, the intention has been to expand on the original format, retaining the concepts that made the initial edition so successful. This is reflected in the structure of this second edition. I am indebted to Professor Doonan for his involvement in this new edition and the continuity that this brings. Each chapter that appeared in the original volume has been reviewed and updated to reflect advances and bring the topic into the 21st century. In many cases, this reflects new applications or new matrices available from vendors. Many of these have increased the performance and/or scope of the given method. Several new chapters have been introduced, including chapters on all the currently used protein fractionation and ch- matographic techniques. They introduce the theory and background for each method, providing lists of the equipment and reagents required for their successful execution, as well as a detailed description of how each is performed.
